A single diffusion technique in which agar containing antiserum is placed in a test tube and antigen is layered over it; precipitin lines form where the concentrations of each antigen and antibody are equivalent.

Le principe de la technique (décrite par Jacques Oudin en 1946) consiste à remplir un tube de verre d'un gel d'agar dans lequel un antisérum a été préalablement incorporé, puis à appliquer une solution d'antigène dans le tube. L'antigène progresse par simple diffusion dans le gel en créant un gradient de concentration. Si la concentration initiale d'antigène est suffisante, un précipité se forme au niveau du front de progression de l'antigène.
